How well do cell signal boosters work in multi-floor houses?

Signal boosters can work successfully in multi-floor homes under certain conditions.

Let’s use, as an example, a two-story home with a 9-foot ceiling upstairs, a 10-foot ceiling downstairs, and 1 foot between floors—20 total feet. If the homeowner mounts a dome antenna on the ceiling of the second floor, and the antenna’s broadcast distance is 50 feet, its signal should cover both floors; right?

If the home has wood joists and carpeted floors, then yes: The upstairs antennas should provide cell signal to both floors. If the home uses concrete between floors, this will almost certainly require antennas on each floor (concrete blocks cellular signals). If the upstairs floor covering is stone or tile, these materials will reduce the signal to the lower floor, which may require additional antennas.
